摘要
目的:观察电针结合鳖甲煎丸对H22肝癌小鼠肿瘤抑制作用及JNK信号通路的影响。方法:建立H22肝癌小鼠模型,将100只肝癌小鼠随机分为模型组、环磷酰胺组、电针组、鳖甲煎丸组、针药联合组,小鼠治疗14 d后,取肿瘤组织称重,计算抑瘤率;采用Western blot方法检测肿瘤组织Bcl-2、Bax、JNK、p-JNK蛋白表达情况。结果:针药联合对H22小鼠肿瘤生长有明显抑制作用;Western blot结果表明针药联合能明显降低Bcl-2蛋白表达,上调Bax蛋白表达,促进JNK蛋白磷酸化。结论:针药联合可通过激活JNK信号通路,降低抑凋亡蛋白Bcl-2表达,提高促凋亡蛋白Bax表达,调整Bcl-2/Bax比值,进而发挥抑瘤作用。
Objective: To observe electro-acupuncture( EA) binding Bie Jia Jian Wan( BJJW) tumor-inhibition on H22 liver cancer mice and influence with JNK signaling pathway. Method: A total of 100 H22 liver cancer mice models were set and then randomized into 5 groups: Model,Cyclophosphamide,EA,BJJW,and EA binding BJJW group. After 14 days of different treatments,then killed the mice and calculate the weight of tumor tissue and measured the inhibition rate. The Bcl-2,Bax,JNK,p-JNK protein expression of tumor tissue were detected by Western Blot assay. Result: Western Blot results showed that EA could significantly reduce Bcl-2 protein expression,increase Bax protein expression and promote protein phpsphorylation of JNK. Conclusion: Evidences show that EA binding drug can exert anti-tumor role by activating the JNK signaling pathway,reducing the expression of anti-apoptosis protein Bcl-2 and improving the apoptotic Bax expression,adjusting the ratio of Bcl-2 / Bax.
