全自动细胞分选仪对单细胞筛选的优化与应用

摘要 以Ec-109、BHK细胞为例,采用AVISO细胞分选仪进行单细胞的分选与优化。首先,将电脑中的细胞筛选软件与显微镜相连接,依据不同的细胞类型,设置合适的挑选参数,并从X、Y与Z轴层面精确校准挑选位置。通过自由设定细胞特征参数,可准确地挑选到目标细胞。最后,检测分析细胞筛选率,均可达到90%以上。该筛选技术对单细胞的挑选可提高到可视化水平,与常规筛选方法相比,样品不需特殊处理,细胞收集方便,节约操作时间,提高了实验效率。 A novel, facilitate and rapid high-throughput technology for cell directional selection was introduced. The operation processes of single cell selection and optimization were detailly described by using Ec-109 cells and BHK cells as examples. Firstly, the screening software in the computer was connected with micrnscope. The appropriate parameters of screening were setting according to the ceil type, and the location of screening was calibrated from the X axis, Y axis and Z axis. The target cells were selected ac, curately through freely setting cell feature parameters. Finally, cell screening rates were detected and analyzed, and the results could reach above 90%. The new automated screening technology of single cell can be achieved to the visual level compared with the conventional screening ,nethod. The sample dnses not need special treatment, and cells are collected conveniently. The new method saves the operating time, and improves the experiment efficiency significantly.
