肝靶向干扰素对小鼠免疫毒性的初步研究 被引量:2

摘要 目的评价肝靶向干扰素(interferon-circumsporozoite protein,IFN-CSP)的免疫毒性作用。方法实验设IFN-CSP低、中、高剂量组(分别为1.87×107IU/kg,5.6×107IU/kg,1.68×108IU/kg),溶剂对照组(生理盐水),阳性对照组(环磷酰胺),小鼠经皮下注射给药2周,迟发型超敏反应检测细胞免疫功能,溶血空斑试验检测体液免疫的功能。结果环磷酰胺阳性对照组与溶剂对照组的小鼠比较,肿胀度、脾指数、胸腺指数明显下降(P<0.05),低、中、高剂量组与溶剂对照组相比,肿胀度、脾指数、胸腺指数没有显著差异。环磷酰胺阳性对照组的空斑数与溶剂对照组相比明显下降(P<0.05),而低、中、高剂量组与溶剂对照组相比,空斑数没有显著差异。结论 IFN-CSP对小鼠的细胞免疫功能及体液免疫功能均无明显影响,这为进一步的安全性评价提供了依据。 Objective To evaluate the immunotoxicity effect of Liver targeting interferon (IFN -CSP) on mice.Methods Mice were randomly divided into five groups:low, middle and high dose of IFN-CSP, solvent control group(saline) and Positive control group (cyclophosphamide).They were injected subcutaneously for 2 weeks.Delayed hypersensitivity test was used to determine the cell immunefunction and plaque forming cell assay was used to determine the humoral immune function.Results There was no significant difference of the the index of immune organ and the ear swelling degree between IFN-CSP groups and control group.There was also no significant difference on hemolytic plaque test between them.Conclusion IFN-CSP has no significant effect on both cellular immunity function and humoral immune function of mice, this results will provides the basis for further safety evaluation.
