摘要
目的观察蒺藜总皂苷对大鼠视网膜神经细胞凋亡的影响。方法成年健康雄性SD大鼠25只,分为对照组,缺血再灌注组和蒺藜总皂苷组,建立视网膜缺血再灌注损伤模型。应用免疫印迹法检测细胞周期素D1(cyclin D1)和细胞周期素依赖性激酶4(CDK4)的表达,采用流式细胞计数法检测细胞凋亡率。结果与对照组比较,缺血再灌注组cyclin D1、CDK4表达升高,凋亡细胞增多(P<0.01);与缺血再灌注组比较,蒺藜总皂苷组cyclin D1、CDK4表达和凋亡细胞明显减少(P<0.05或0.01)。结论蒺藜总皂苷对视网膜缺血再灌注损伤有拮抗作用,其机制可能与其调控细胞周期因子从而抑制细胞凋亡有关。
Objective To investigate the effects of gross saponin from Tribulus terrestris L ( GSTT) on neuronal apoptosis in retina after ischemia reperfusion in rats .Methods 25 adult male Sprague-Dawley rats were randomly divided into control ,ischemia and GSTT groups .A model of ischemia reperfusion in rats was es-tablished .Western blot and flow cytometry were used respectively to observe the expression of cyclin D 1 and CDK4 protein and neuronal apoptosis rate in retina .Results A significant increase in cyclin D 1 and CDK4 ex-pression was observed after reperfusion .The differences of cyclin D1,CDK4 and cell apoptosis between ischemia group and GSTT group were significant (P〈0.05 or P〈0.01).Conclusion GSTT can protect retina by regu-lating cell cycle factors and inhibiting cell apoptosis in ischemia reperfusion injury .
