HPLC法测定壮骨胶囊中补骨脂和异补骨脂素的含量

Content determination of Psoralen and Isopsoralen in Zhuanggu capsules by HPLC

目的：建立高效液相色谱测定壮骨胶囊中补骨脂素和异补骨脂素含量的方法。方法色谱柱： C18（4.6＆#215;250 mm，5μm）；流动相：30％乙腈－0.5％乙酸水；流速：1.0 ml／min；柱温：25℃；检测波长：246nm；进样量：20μl。结果补骨脂素在0.002～0.04 mg／ml呈良好线性关系（r＝0.9997，n＝5），平均回收率为95.28％，RSD为1.84％（n＝5）；异补骨脂素在0.002～0.04 mg／ml呈良好线性关系（r＝0.9999，n＝5），平均回收率为96.37％，RSD为1.76％（n＝5）。结论本法操作简便，准确，灵敏，可用于壮骨胶囊补骨脂素和异补骨脂素的含量测定。

Objective To establish a method of high-performance liquid chromatography for detremining the contents of Psoralen and Isopsoralen in Zhuanggu capsules .Methods Column：C18（4.6 ＆#215;250 mm,5 μm,）;mobilephase：30%CH3CN-water（0.5%CH3COOH）;flow：1.0 ml/min;temperature：25℃;detection：246nm;sample volume：20 μl.Results The Psoralen had a good linearity in 5.4~54 μg/ml （r=0.9995,n=5）, the average recovery was 97.54%（RSD=1.84%,n=5）, The Isopsoralen had a good linearity in 5.2~52μg/ml （r=0.9999,n =5）, the average recovery was 97.40% （RSD =1.76%,n =5）.Conclusion The method is simple accurate and sensitive,and can be used for the content determination of Psoralen and Isopsoralen in Zhuanggu capsules .
