摘要 目的牛肾传代细胞系(mardin-darby bovine kidney,MDBK)通常用于疫苗生产过程中病毒的增殖,为了检验该细胞系安全性、监测细胞培养过程中是否发生染色体畸变,确保细胞系有清楚的遗传背景。对MDBK细胞系的原始细胞库、基础细胞库、工作细胞库最高限制代次细胞库以及超出最高限定代次10代的细胞进行了细胞核型分析。方法通过对不同代次细胞的染色体核型对比数据及染色体模式数变异率的分析数据,包括每个代次细胞染色体相对长度和着丝粒指数、染色体众数、染色体核型模式图等的数据的对比分析,结果该细胞系染色体分布频率在正常二倍体细胞(2n=52)附近。结论该细胞系在使用代次内染色体无显著变异,属于优质细胞系,可用于疫苗生产中病毒的增殖,使用代次范围控制在工作细胞库最高限制代次以内。 Objective: MDBK (mardin-darby bovine kidney) are typically used during proliferation of the virus for vaccine production, in order to test the safety of ceils, monitoring whether the cell of chromosomal aberration during the culture, and ensure a clear genetic background of the cell. Defined the cell karyotype analysis experiments of the MDBK cells in the original cell line library, the basic cell bank, working cell bank generation in the maximum limit of the cell bank and passages 10 above the maximum. Methods: Comparative data analysis of variance rate data through the different generations of cells karyotype and chromosome pattern, including the relative length of each chromosome generations and centromere index, data chromosome number, karyotype pattern diagrams of comparative analysis. Results: The result showed that the frequency distribution of the cells in the vicinity of chromosomes normal diploid cells (2n=52) . Conclusion: the use of cell lines in generations was no significant variation in the chromosome, a quality cell lines can be used for vaccine production in the proliferation of the virus, using passages controlled within the range of the maximum limit of working cell bank generations.
