CD51阳性心肌细胞对心肌梗死后小鼠心功能改善的作用 被引量:3

摘要 目的探讨CD51阳性(CD51+)的心肌细胞移植治疗AMI后心脏结构及功能变化。方法取C57小鼠14只随机分为对照组(n=7)和细胞移植组(n=7),结扎冠状动脉左前降支制成心肌梗死模型。细胞移植组心肌梗死后立即于心肌梗死部位注射转染红色荧光蛋白的CD51+心肌细胞(RFP-CD51),对照组注射生理盐水。移植后1周及1个月分别用小动物心脏彩色多普勒超声(彩超)仪检测心功能指标的变化及组织形态改变,并且在移植后1个月时观察RFP-CD51细胞是否分化为成熟心肌细胞。结果细胞移植组的小鼠在1周及1个月时的心脏彩超均显示LVEF、左心室缩短分数(LVFS)均显著高于对照组(P<0.01),左心室收缩末容积(LVESV)小于对照组(P<0.05),而左心室舒张末容积(LVEDV)则无显著差异(P>0.05)。在心肌梗死3周后发现移植的CD51+细胞分化为成熟心肌细胞。结论 AMI后的CD51+心肌细胞移植可分化为成熟心肌细胞、改善心功能、防止心脏重构、减轻心腔扩大。 Objective To discuss the cardiac structural and functional changes after CD51 + myocar-dial cell transplantation in the treatment of acute myocardial infarction (AMI).Methods Fourteen C57 mice were randomly divided into the control (n =7)and cell transplantation groups (n =7).Left anterior descend-ing coronary artery was ligated to establish AMI mouse model.In the cell transplantation group,CD51 + myo-cardial cells transfected with red fluorescent protein (RFP-CD51 )were injected to the myocardial infarction sites immediately after AMI,and phosphate buffer saline was administered in the control group.At 1 week and 1 month after cell transplantation,the changes in the heart function indexes and tissue morphology were detec-ted by small animal ultrasonography.The differentiation of RFP-CD51 cells into mature myocardial cells was observed at 1 month following cell transplantation.Results Small animal ultrasonography revealed that left ventricular ejection fraction (LVEF)and left ventricular fractional shortening (LVFS)were significantly higher compared with those in the control group at 1 week and 1 month after cell transplantation (both P 〈0.01 ).Left ventricular end-systolic volume (LVESV)in the cell transplantation group was significantly lower than that in the control group (P 〈0.05).Left ventricular end-diastolic volume (LVEDV)did not significantly differ be-tween two groups (P 〉0.05).The transplanted CD51 + cells were differentiated into mature myocardial cells at 3 weeks after AMI.Conclusions After AMI,transplanted CD51 + myocardial cells can differentiate into ma-ture myocardial cells,which improve heart function,prevent cardiac remodeling and mitigates cardiac chamber enlargement.
