转MdSPDS1-AtCBF1双价基因烟草抗逆境研究

摘要 为研究由诱导型启动子Atrd29A介导表达的2个抗胁迫基因MdSPDS1和AtCBF1对转化烟草的影响,对转化双价基因烟草进行了抗逆境试验,主要包括:耐盐性试验、耐渗透性试验和耐寒性试验。结果表明:转基因烟草在添加了20μmol/L Al2(SO4)3、20μmol/L CuSO4、100mmol/L NaCl的培养基上生长时,质量和高度的增加率都大于未转基因烟草,表现出一定的抗盐性。同样,在添加了50mmol/L Mannitol、4mmol/L PEG的培养基上,转基因烟草也得到了比未转基因烟草更好的生长状态。对转基因烟草组培苗进行-4℃耐寒性试验,具有比未转基因烟草更低的细胞死亡和超氧阴离子累积。移栽到温室中的转基因烟草在-4℃处理时叶片萎蔫减缓,相对电导率低于未转基因烟草。 In order to discuss the effect of MdSPDS1-AtCBF1 double genes mediated by inducible promoter Atrd29A on transgenic tobacco, two transgenic tobacco lines and a control strain were taken as materials for studying stress resistance including tests for salt tolerance, osmotic tolerance and cold tolerance. The results indicated that the transgenic lines showed a significant increase in the increment percentage of shoot height and weight compared with the wild-type in the medium containing 20 μmol/L Al2 (SO4)3,20 μmol/L CuSO4,100 mmol/L NaCI,50 mmol/L Mannitol,4 mmol/L PEG respectively. Treatment at - 4 13 showed that the transgenic tissue culture lines exhibited better cold tolerance. This might due to that the cell death and accumulation of superoxide anion were inhibited in the transgenic plants. Under this condition,the transgenic tobaccos transferred in greenhouse showed a tendency of slow wilting with lower relative conductivity.
