Determination of platycodin in Platycodi Radix from different habitats

目的建立测定桔梗Platycodon grandiflorum中桔梗总皂苷和桔梗皂苷D的测定方法,并准确测定我国不同产地桔梗药材中桔梗总皂苷和桔梗皂苷D的量。方法采用超声波甲醇提取法,称重法测定桔梗总皂苷量;采用HPLC法测定桔梗皂苷D量,色谱条件:Waters Symmetry-C18色谱柱,流动相为乙腈-水(22∶78),体积流量1.0 m L/min,检测波长为210 nm,柱温为30℃。结果不同产地的桔梗药材中桔梗总皂苷和桔梗皂苷D量差别较大。在所测的桔梗药材中,浙江产桔梗总皂苷量最高,为12.03%;黑龙江、辽宁、安徽、山东、江西产桔梗总皂苷量均高于《中国药典》2005年版规定的6%;而其余各省的量均低于6%,最低的为河南,其量仅为1.57%。对于桔梗皂苷D来说,云南产桔梗的量最高,为0.654%;河北产桔梗的量最低,为0.017%。结论建立的方法操作简便,测定结果准确可靠,可用于桔梗药材中桔梗总皂苷和桔梗皂苷D量的测定。在我国不同产地桔梗药材中,桔梗总皂苷和桔梗皂苷D的量存在差异。同一药材中桔梗总皂苷与桔梗皂苷D的量,未显示出相关性。

Objective To establish a method of the determination of total platycodin and platycodin D; And to determine the contents of total platycodin and platycodin D in Platycodi Radi X from different habitats. Methods The content of total platycodin was determined by 1ultrasonic extraction and weight method; And the content of platycodin D was determined by HPLC. The chromatographic conditions were Waters Symmetry-C18 column, mobile phrase of CH3CN-H2O（22：78）, flow rate of 1.0 m L/min, detection wavelength at 210 nm and column temperature 30 ℃. Results There was a greater difference between the contents of total platycodin and platycodin D in Platycodi Radix from different habitats. In measuring Platycodi Radix, the content of total platycodin in Platycodi Radix from Zhejiang Province was the highest, which was 12.03%; the contents in Platycodi Radix from Heilongjiang, Liaoning, Anhui, Shandong, and Jiangxi Provinces were more than 6%; However, the contents in Platycodi Radix from the other provinces were lower than 6%, and it was the lowest in Platycodi Radix from Henan province, which was 1.57%. The contents of platycodin D in Platycodi Radix from Yunnan province was the highest, and it was the lowest in Platycodi Radix from Hebei province. Conclusion The methods could be used for the determination of total platycodin and platycodin D in Platycodi Radix because of theirs simple operations with accurate and reliable results. The contents of total platycodin and platycodin D are different obviously in Platycodi Radix from different habitats, and the contents of total platycodin and platycodin D in the same drug with the level of inconsistency don＇t show correlation.
