一株假芝的分离鉴定与生物学特性 被引量:15

Isolation, identification, and biological characteristics of a wild Amauroderma mushroom

摘要 灵芝是我国著名传统药用真菌,假芝属(Amauroderma)真菌作为灵芝科重要属,我国已报道种共有22个.本研究从广州白云山采集到一株野生假芝子实体,经组织分离获得菌丝体纯培养,编号GZ-01菌株.利用形态鉴定和ITS鉴定手段,确定其分类学地位为假芝属,系统命名为A.subrugosum,为中国首次记录种,菌种保藏号ACCC52954.生物学特性研究表明,菌丝生长的最适碳源为甘露醇,最适氮源为酵母浸膏或大豆蛋白胨,最适C/N比为20/1,最适生长因子为维生素B6,最适生长温度为28℃,最适p H值范围为6.0-7.5.本研究获得新的药用真菌资源,其培养条件研究可为进一步开发利用奠定实验基础. Reishi mushrooms are Chinese traditional medicinal fungi with antioxidant, antitumor, immunomodulatory activities. Amauroderma is one of the important genera in the family Ganodermataceae. All together 22 Amauroderrna species have been reported in China. This study isolated a pure cultured strain GZ-01 from mushroom fruiting bodies collected in the Baiyun Mountain, Guangzhou Province. Based on morphological and ITS identification methods, it was identified as Amauroderma subrugosum with the collection No. of ACCC52954. It is a new record of the genus Amauroderma in China based on morphological and molecular identification. Culture conditions study showed the optimum condition for the myceliao growth included mannitol as the carbon source, yeast extract or soy peptone as nitrogen source, the C/N rate as 20/1, growth factor of Vitamin B6. temperature of 28℃, and the pH range as 6.0-7.5. The results show that the fungus is a newly reported medicinal fungal species, which is an important reference for further artificial domestic cultivation and applications of the mushroom resource.
