FC/TK对大鼠卵巢癌的免疫治疗作用的实验研究

摘要 目的:探讨自杀性肿瘤疫苗(FC/TK)对大鼠卵巢癌的治疗作用。方法:采用聚乙二醇法,将自然杀伤基因修饰的卵巢癌细胞与树突状细胞融合制备成FC/TK,选取40只雌性Fischer344大鼠,制备Fischer344大鼠卵巢癌荷瘤模型,采用随机数字表法分为FC/TK组(FC/TK处理)、未经自然杀伤基因修饰的卵巢癌细胞与树突状细胞融合的疫苗(FC)处理组(FC组)、经60Co照射的FC/TK处理组(60Co-FC/TK组)和对照组(等量PBS处理)各10只。计算各组大鼠90天后的肿瘤平均重量、抑瘤率等指标。结果:FC组、FC/TK组的瘤体重量(3.17±0.68)g、(2.92±0.59)g均显著低于60Co-FC/TK组和PBS组且差异均具有统计学意义(P<0.05);FC组、FC/TK组的抑瘤率(61.53±10.28)%、(64.56±11.77)%显著高于60Co-FC/TK组且差异具有统计学意义(P<0.05)。FC组、FC/TK组的血清IL-2、TNF-ɑ水平显著低于60Co-FC/TK组和PBS组且差异均具有统计学意义(P<0.05)。FC组、FC/TK组的瘤体实质中CD4+、CD4+/CD8+比例显著的高于60Co-FC/TK组和PBS组,CD8+细胞显著的低于60Co-FC/TK组和PBS组且差异均具有统计学意义(P<0.05)。结论:FC/TK对大鼠卵巢癌具有显著的治疗作用,可能和提高肿瘤实质内部的T淋巴细胞亚群水平、降低血清炎症因子水平有关。 Objective: To investigate the Dutch act tumor vaccine (FC/TK) treatment effect on rat ovarian cancer. Methods: U- sing poly (ethylene glycol), natural killer gene modified ovarian cancer cells with dendritic cells were prepared by the fusion of a FC/TK. 40 female Fischer 344 rats were selected to prepare Fischer344 rats ovarian cancer tumor model and were randomly divided into FC/TK group (FC/TK processing), the suicide gene modified ovarian cancer cells with dendritic cell fusion vaccine (FC) processing group (FC group), the FC/TK 60Co irradiation treatment group (60Co-FC/TK group) and control group (the same amount of PBS treatment), 10 rats in each. After 90 days, the average tumor weight, inhibition rate and other indicators were calculated. Results: Tumor weight of FC group and FC/TK group was (3. 17±0. 68) g, (292± 0. 59 ) g, which were significantly lower than that of 60Co-FC/TK group and PBS group and the difference was statistically significant (P〈0. 05). The tumor inhibition rate of FC group and FC/TK group was (61.53±10. 28)%, (64. 56± 11.77)% , which was higher than that of 60Co-FC/TK group and the difference was significant. Serum IL-2 and TNF alpha levels of the FC group and FC/TK group were significantly lower than those of 60Co-FC/TK group and PBS group and the differences were statisti- cally significant (P〈0. 05) . CD4 + and ratio of CD4 + / CD8 + in tumor essence of FC group and FC/TK group were higher than those of 60Co-FC/TK group and PBS group, CD8 + cells of FC group and FC/TK group were significantly lower than those of 60Co-FC/TK group and PBS group and the difference was statistically significant (P〈O. 05 ) . Conclusion: FC/TK on rat ovarian cancer has significant thera- peutic effect and may improve tumor parenchyma internal T lymphocyte subsets level, reduce the serum levels of inflammatory cytokines. [
