miR-142-3p对小鼠泌乳量、乳成分及乳腺组织结构的影响 被引量:2

摘要 MicroRNA(miRNA)是一类长度约18~25个核苷酸序列的非编码小分子RNA,miR-142-3p能通过调控靶基因表达,来调控乳腺发育和泌乳。通过尾静脉注射实验技术,改变miR-142-3p在小鼠乳腺组织中的表达量,观察记录48h内泌乳量的变化,使用乳成分分析仪、光镜技术检测和观察miR-142-3p对小鼠乳汁成分和乳腺组织结构的影响。结果表明,miR-142-3p过表达后,泌乳量降低、乳品质下降、泌乳期乳腺组织中腺泡数量减少,腺泡体积变小。miR-142-3p沉寂后,泌乳量增加、乳品质提高、泌乳期乳腺组织中腺泡数量增加,腺泡体积增大。研究表明,miR-142-3p对乳腺发育、泌乳以及乳品质具有负向调控作用。 MicroRNAs(miRNAs) are a kind of non-coding small RNA of about 18 to 25 nucleotides in length. MiR-142-3p can regulate mammary gland development and lactation through the regulation of target gene expression. In this test, tail vein injection techniques were used to changed the expression of miR-142-3p in mouse mammary tissues. The change of milk yield was observed and recorded within 48 hours. The effect of miR-142-3p on the mouse milk composition and the structure of mammary tissues were analyzed by the milk composi- tion fast analyzer and the light microscope .The results showed that when miR-142-3p was over expressed ,the milk yield reduced, the quantity of the milk was down-regulated, the mammary acinus of mouse lactating mammary gland reduced, the acinus shrink. When miR-142-3p was inhibited, the milk yield increased, the quantity of the milk was up-regulated, the mammary acinus of mouse lactating mammary gland increased, and the acinus expand. The results suggest that miR-142-3p has a down- regulation function on the mammary gland development, lactation and milk quality.
