摘要
目的:观察小半夏汤对化疗大鼠胃肠激素水平的影响,以探讨其防治化疗性恶心呕吐的机制。方法:雄性Wistar大鼠60只,随机分为空白对照组、小半夏汤正常对照组、顺铂模型组、昂丹司琼组、小半夏汤大剂量组和小剂量组。除前两组腹腔注射等容量生理盐水外,其余各组腹腔注射顺铂6 mg/kg建立化疗性异食癖模型。造模前1 h及造模后11 h分别灌胃给药,每次各组均以10 m L/kg容积给以蒸馏水、小半夏汤0.8 g/kg、蒸馏水、昂丹司琼1.3 mg/kg、小半夏汤1.6 g/kg、小半夏汤0.8 g/kg。每12 h记录动物高岭土摄入量。造模24 h后取血,酶联免疫法检测血清胃肠激素水平。结果:模型组动物摄食高岭土量显著升高,外周血中胃泌素和胃动素水平降低、生长抑素与血管活性肠肽升高。小半夏汤在减少模型动物摄食高岭土的同时,可升高胃泌素和胃动素水平(P<0.05)、降低生长抑素与血管活性肠肽水平(P<0.05)。结论:小半夏汤防治化疗性恶心呕吐作用机制与其改善胃肠激素紊乱有关。
Objective:In order to explore the anti- emetic mechanism of Xiaobanxia decoction(XBXT),its effect on gastrointestinal hormone level was observed in chemotherapy- induced pica rats. Methods:Sixty male Wistar rats were randomly divided into normal control,XBXT control,cisplatin model,ondansetron,XBXT high dose and low dose groups. Pica model was induced by cisplatin 6 mg/kg i.p.. One hour before and 11 hour after cisplatin injection,each group was gavaged with distilled water,XBXT 0.8 g/kg,ondansetron 1.3 mg/kg,distilled water,XBXT 1.6 g/kg and 0.8 g/kg respectively. After 24 h of cisplatin injection,gastrointestinal hormones in serum were measured using ELISA method. Results:Both dosages of XBXT could significantly inhibit rat pica(P0.01),and reverse the abnormal increase of somatostatin and VIP(P〈0.05) and abnormal decrease of gastrin and motilin(P〈0.05). Conclusion:The mechanism of XBXT on the treatment of chemotherapy- induced nausea and vomiting was related to ameliorate the disorders of gastrointestinal hormone.
