摘要
目的:探讨黄芪多糖对大鼠脑缺血再灌注损伤后脑组织SOD、MDA及脑指数的影响。方法:SPF级Wistar大鼠72只,分为假手术组、模型组、阳性对照组、黄芪多糖高剂量组、黄芪多糖中剂量组、黄芪多糖小剂量组,采用改良的线栓法制作大鼠脑缺血再灌注模型,各药物组分别给予相应药物15天后,检测各组大鼠脑指数、黄嘌呤氧化酶法测定SOD的活力、硫代巴比妥酸(TBA)法检测MDA含量的表达。结果:1与假手术组比较,模型对照组大鼠脑组织中的SOD水平明显降低;与模型对照组比较,除黄芪多糖低剂量组差异无统计学意义外,其余各组差异均有统计学意义。2与假手术组比较,模型组大鼠脑组织中的MDA水平明显升高;与模型组比较,各药物组MDA含量均降低,黄芪多糖高剂量组与模型组比较,差异有统计学意义。3与假手术组比较,模型组大鼠脑指数差异有统计学意义,其余各组与模型对照组相比,呈下降趋势,但差异无统计学意义。结论:黄芪多糖改善脑缺血再灌注的作用机制与提高脑缺血再灌注模型大鼠SOD水平,降低MDA含量,改善脑指数有关。
Objective: To explore the effects of astragalus polysaccharides on SOD, MDA content of brain and brain index in cerebral ischemia reperfusion rats. Methods: The SPF level of 72 wistar rats were divided into sham operation group, model group, positive control group, astragalus polysaecharide high dose group, astragalus polysaceharide middle dose group, astragalus polysaccharide small dose group. Cerebral ischemia reperfusion model rats were made through reformative thread embolism method. After the medicine groups were given medication for 15 days, the brain index, and expression of SOD by xanthine oxidase assay, MDA by thiobarbituric acid method were detected in each group. Results: (1)Compared with the sham operation group, the expression of SOD reduced significantly in the model group, compared with the model group, other groups had significant difference except low dose of astragalus polysaccharides. (2)Compared with the sham operation group, MDA level of model group rats in the brain tissue reduced significantly (P〈0.05). Compared with the model group, MDA content of the drug group decreased, astragalus polysaccharides high dose group had statistical significance. (3)Compared with the sham operation group, brain index had significant difference in the model group. Compared with the model group, the other groups had the downward trend but there was no significant difference. Conclusions: Astragalus polysaccharides can improve action mechanism of the reperfusion in cerebral ischemia rats,which may be due to increase the SOD level, decrease the content of MDA, improve the brain index.
