摘要
目的探讨蛋白酪氨酸磷酸酶14(PTPN14)对乳腺癌细胞增殖能力的影响。方法首先在乳腺癌细胞株MCF-7中建立稳定高表达PTPN14的细胞系,并通过RT-PCR和蛋白免疫印迹实验进行鉴定;然后通过RNA干扰技术靶向抑制蛋白表达,采用细胞计数法、噻唑蓝(MTT)法、细胞克隆形成实验、BrdU细胞掺入实验以及裸鼠体外成瘤实验,检测PTPN14对乳腺癌细胞MCF-7细胞增殖的影响。结果通过在mRNA和蛋白水平的检测,证实高表达PTPN14的MCF-7稳定细胞系建立成功。MTT法结果表明,培养第5天时高表达PTPN14组的MCF-7细胞数约为对照组的60%(P<0.05);细胞计数实验表明,与对照组比较,高表达PTPN14的MCF-7细胞数在培养第3、4、5天时明显减少(P<0.05);细胞克隆形成实验中,对照组和高表达PTPN14组的克隆数分别为(125±25)个和(62±13)个,差异具有统计学意义(P<0.05);同时,低表达内源性PTPN14将明显促进细胞增殖。进一步的裸鼠体内成瘤实验中,高表达PTPN14组的肿瘤体积仅为对照组的1/5(P<0.05);对照组和PTPN14组的肿瘤重量分别为:(0.6±0.2)g和(0.2±0.1)g,差异具有统计学意义(P<0.05)。结论高表达PTPN14抑制乳腺癌细胞的增殖能力。
Objective To explore the effect of PTPN14 on the cell proliferation of breast cancer cells.Methods The PTPN14-overexpressing MCF-7cell line was established and confirmed by using RT-PCR and Western blot.Then the effect of PTPN14 on the proliferation of MCF-7cells was evaluated by cell counting,MTT,colony formation,BrdU incorporation assay and xenograft tumor formation assay after inhibition of PTPN14 protein expression with RNA interference technology.Results The MCF-7stable cell line overexpressing PTPN14 were successfully established by confirming the high expression of PTPN14 at mRNA and protein level.MTT assay showed that the cell number was decreased by at least 40% at the fifth day in PTPN14 overexpression group compared to control group(P〈0.05).Cell counting showed that the cell number was significantly decreased at3,4and 5days of culture in PTPN14 overexpession group compared with control group(P〈0.05).In the colony formation assay,the colony number was(125±25)in control group and(62±13)in PTPN14 overexpression group,with difference being statistically significant(P〈0.05).On the contrary,knockdown of endogenous PTPN14 promoted the proliferation of MCF-7cells.In the xenograft tumor formation assay,the xenograft weight was(0.2±0.1)g in PTPN14 overexpression group,significantly lower than in control group[(0.6±0.2)g,P〈0.05].Conclusion Overexpression of PTPN14 can inhibit the proliferation of breast cancer cells.
