摘要
目的:建立南五味子中核苷类成分的分析方法,对其核苷类成分进行分析评价,为南五味子资源的综合利用提供科学依据。方法:采用UPLC-TQ/MS技术分析测定9个产地、70批南五味子样品中16种核苷类成分的含量。使用ACQUITY UPLC BEH Amide色谱柱(100 mm×2.1 mm,1.7μm),以10 mmol·L-1乙酸胺+0.8%乙酸的水溶液(A)-0.05%乙酸的乙腈溶液(B)为流动相,梯度洗脱(0-6 min,10%A;6-8 min,10%A→40%A;8-9 min,40%A;9-11 min,40%A→50%A),流速0.4m L·min-1,柱温35℃。多反应监测(MRM)模式。结果:南五味子中含有多种核碱基、核苷和核苷酸类成分,所测成分中含量最高的是尿苷,含量最低的是2'-脱氧鸟苷。不同产地间核苷种类几无差异,不同产地间含量差异最大的成分是腺苷,各成分含量差异可能与地域有关。结论:建立了准确高效分析评价南五味子中核苷类成分的方法,研究结果可为南五味子功效物质基础的进一步研究和其资源开发利用提供科学依据。
Objective:To establish an analytical method of nucleoside compounds in Schisandrae Sphenantherae Fructus,evaluate and analyze the nucleoside compounds in Schisandrae Sphenantherae Fructus,so as to provide a scientific basis for the comprehensive utilization of resources.Methods:The UPLC-TQ/MS method was used to analyze and measure 16 kinds of nucleoside compounds in 70 batches of Schisandrae Sphenantherae Fructus from 9places of origin.An ACQUITY UPLC BEH Amide C18column(100 mm × 2.1 mm,1.7 μm) was used,with a mobile phase consisting of A(0.8% acetic acid and 10 mmol·L-1ammonium acetate in aqueous solution) and B(0.05%acetic acid in acetonitrile).UPLC linear gradient conditions were:0-6 min,10% A;6-8 min,10% A→40% A;8-9min,40% A;9-11 min,40% A→50% A.The injection volume was 0.4 m L·min-1,and the column temperature was maintained at 35 ℃ under negative ionization(MRM) mode.Results:Schisandrae Sphenantherae Fructus contained a variety of ingredients of nucleobases,nucleosides and nucleotides,among with the highest concentration of the measured compounds was uridine while the lowest content was 2'-deoxyguanosine.There was no big difference between different types of non-nucleosides.The greatest difference in content of ingredients of different origins was seen in adenosine,and geographical differences in the content of each compound were relevant.Conclusion:The method for analysis of nucleosides in Schisandrae Sphenantherae Fructus can better evaluate the nucleoside compounds in Schisandrae Sphenantherae Fructus.The experimental results provide a scientific basis for the further research and for the development and utilization of Schisandrae Sphenantherae Fructus.
