New technology of enzymic extraction red pigment from black soybean skin with microbial fermentation crude enzyme liquid

摘要 通过微生物液体发酵技术得到富含纤维素酶的发酵粗酶液,对黑豆衣进行酶解处理,以红色素提取率为指标,确定微生物发酵粗酶液酶解提取最佳工艺条件。从自然界筛选出一株高产纤维素酶菌株w8,确定其最佳产酶发酵条件为:初始p H值为8、1%玉米芯粉、0.5%硫酸铵、0.05%吐温80、温度为30℃、接种量为3%、培养5 d,优化后,其FPA酶活提高至510.19 U/m L,CMC酶活提高至3879.16 U/m L。微生物发酵粗酶液酶解提取黑豆衣红色素最佳条件为:料液比为1:20、酶解温度为50℃、酶解时间120 min、酶用量为50 U/m L、初始p H值为6,浸提4次,合并滤液经真空冷冻干燥获得黑豆衣红色素,提取率达到96.84%。 Rich in cellulose enzyme is obtained by liquid fermentation technology of crude enzyme liquid fermentation, enzymatic hydrolysis was carried out on the black soybean skin processing, red pigment extraction rate as the index, to determine the microorganism fermentation crude enzyme liquid enzymic extraction optimum technological conditions. Screening of high-yield cellulase strains w8 from nature. Determine the optimal fermentation conditions for producing enzyme: initial pH of 8, 1% corn powder, 0.5% ammonium sulfate, 0.05% tween 80, temperature of 30 %, 3% inoculated quantity and cultivate 5 d, after optimization, the FPA enzyme activity increased to 510.19 U/mL, CMC enzyme activity increased to 3879.16 U/mL. Microorganism fermentation crude enzyme liquid enzymic extraction conditions of the black soybean skin red pigment as extracting agent for material liquid ratio of 1:20, enzymolysis temperature of 50 %, enzyme dosage 50 U/mL, enzymolysis time of 120 rain, initial pH value of 6, leaching four times, merge the filtrate obtained with vacuum freeze drying black soybean skin red pigment, extraction rate reached 96.84%.
