环巴胺抑制Shh信号对乳腺癌细胞增殖和凋亡的影响 被引量:2

摘要 目的探讨环巴胺靶向抑制Shh信号对乳腺癌细胞增殖、凋亡以及Survivin表达的影响。方法采用荧光定量PCR法检测乳腺癌组织及癌旁正常组织中Shh、PTCH1及GLi1的表达,MTT法检测不同浓度环巴胺对人乳腺癌细胞MCF-7增殖抑制的影响,流式细胞术检测环巴胺对MCF-7细胞凋亡的影响,Western blot检测环巴胺处理前后MCF-7细胞GLi1和Survivin的表达。结果 Shh、PTCH1及GLi1在乳腺癌组织中均呈高表达;不同浓度环巴胺均可抑制MCF-7细胞增殖并诱导其凋亡;环巴胺处理后GLi1和Survivin的表达均降低。结论 Shh信号在乳腺癌组织中呈激活状态,靶向抑制Shh信号可抑制乳腺癌细胞增殖并诱导其凋亡,其可能通过抑制Survivin表达发挥作用。
