RELATIONSHIP BETWEEN GENE TLR9-1486 AND TLR9-2848 POLYMORPHISMS AND EBVAGC SUSCEPTIBILITY

摘要 目的探讨Toll样受体9(TLR9)编码基因TLR9-1486和TLR9-2848位点多态性以及EB病毒(EBV)感染与胃癌易感性的关系。方法选取131例胃癌病人为研究对象,包括43例EBV相关胃癌(EBVaGC)和88例EBV阴性胃癌(EBVnGC),同时随机抽取66例健康人外周血标本作为对照。采用聚合酶链式反应-限制性片段长度多态性(PCR-RFLP)技术检测TLR9基因-1486位点T/C和-2848位点G/A多态性。结果 TLR9-1486T/C位点多态性分析显示,胃癌组和对照组3种基因型分布和等位基因频率比较差异无显著性(P>0.05);EBVaGC组和EBVnGC组中各基因型分布和等位基因频率差异也无显著性(P>0.05);与携带T等位基因者相比较,C等位基因携带者胃癌和EBVaGC发病风险无明显升高趋势。TLR9-2848G/A位点多态性分析显示,胃癌组和对照组3种基因型分布和等位基因频率差异无显著性(P>0.05);EBVaGC组和EBVnGC组中各基因型分布和等位基因频率差异也无显著性(P>0.05);与G等位基因携带者比较,A等位基因携带者胃癌和EBVaGC易感性无明显升高。结论 TLR9-1486和TLR9-2848位点多态性与胃癌易感性无关,且在EBVaGC发生中与EBV感染无协同关系。 Objective To investigate the relationship between toll-like receptor 9 encoding gene TLRg-1486 and TLRg- 2848 site polymorphisms and EBV infection and gastric cancer susceptibility. Methods This study consisted of 131 patients with gastric cancer including 43 cases of EBV-associated gastric carcinoma (EBVaGC) and 88 cases of EBV-negative gastric carcinoma (EBVnGC), and periphera blood specimens taken from 66 healthy individuals served as controls, TLR9 gene 1486 site T/C and -2848 site G/A polymorphism were detected by PCR-restriction fragment length polymorphism analysis (RFLP). Results TLRg-1486T/C site polymorphism analysis showed there was no significant difference in genotype frequency and allele frequency in gastric cancer group and the control group (P2〉0,05), There was also no significant difference in each of genotype distribution and allele frequency between EBVaGC group and EBVnGC group (P〉0.05). Compared with T allele, the frequency of C allele had no apparently increased risk trend for gastric cancer and EBVaGC. TLR9-2848 site G/A polymorphism analysis showed that there was no significant difference in genotype distribution and allele frequency hetween gastric cancer group and the control group (P 2〉 0.05), and there was also no significant difference in each of genotype distribution and allele frequency between EBVaGC group and EBVnGC group (P 2〉 0.05). Compared with G allele, the frequency of A allele showed no obvious risk for gastric cancer and EBVaGC susceptibility. Conclusion TLR9 gene-1486 and-2848 site polymorphism are not associated with gastric cancer, and there is no collaborative relationship with EBV infection in the carcinogenesis of EBVaGC.
