摘要
目的研究碘过量对大鼠膝关节软骨的损伤作用。方法选取1月龄Wistar大鼠随机分为5组进行高碘饮水干预,分别于干预1、5、9月时测定24小时尿碘含量;干预6月和12月时分别处死大鼠,测定血清游离三碘甲腺原氨酸(FT3)、游离甲状腺素(FT4)及促甲状腺激素(s TSH);HE染色观察关节软骨的病理改变。结果高碘摄入后,大鼠尿碘水平成倍增加,100倍高碘组(100HI组)增加尤为明显。干预6个月时,各高碘(HI)组血清s TSH水平较适碘(NI)组有升高趋势。12月时,各HI组雄性大鼠血清s TSH水平较NI组有不同程度的升高,FT4水平出现不同程度的下降。6月时,与NI组相比,各高碘组关节软骨出现退变,表现为各层软骨细胞排列紊乱,潮线断裂、不清晰。至12月时,高碘组出现软骨表面不平,其中100HI组软骨出现严重退变。结论长期摄入过量碘,可导致Wistar大鼠甲状腺功能逐渐衰退,引起关节软骨发生病理学改变。
Objective To explore the influence of iodine excess on thyroid function and articular cartilage. Methods Wistar rats, one month after weaning, were randomly divided into 5 groups. Urinary iodine content was analyzed at the end of 1st, 5th and 9th month. Results Urine iodine level in high iodine(HI) group increased significantly compared with NI group; At 6 months, serum s TSH levels showed a rising trend in HI group; At 12 months, serum s TSH level rised, but FT4 level reduced in male rats in HI group. Compared with NI group, articular cartilage showed disarrangement of chondrocytes, narrowed proliferating cells layer and reduced number of cells in HI group. At 12 months,HI group displayed more serious degenerative. Conclusion Due to long-term ecessive iodine, thyroid function is degraded gradually, resulting in pathologic changes of rat's articular cartilage.
