脐带间充质干细胞辅助游离脂肪的移植研究

摘要 研究脐带间充质干细胞是否能够有效辅助游离脂肪移植。应用脐带间充质干细胞与自体脂肪组织颗粒混合后,移植到裸鼠皮下,与单纯脂肪组织移植作对照,不同的时间观察其各组移植重量,取下后立即固定,包埋、切片,HE染色,观察组织内细胞成活及血管生成情况,通过real-time PCR技术进行检测移植物中的VEGF、PPAR-γ基因表达水平。脐带间充质干细胞可以提高移植物重量及VEGF基因表达水平,但在术后一周、一月、两月观测点与对照组相比差异均没有统计学意义。脐带间充质干细胞不能有效提高移植物重量、成脂以及血管生成的修复效应,不能有效减少术后移植脂肪的吸收,目前应用于游离脂肪移植尚不成熟。 To study the effect of supplement umbilical cord mesenchymal cells (UCMSCs)on adipose tissue transplantation.We used supplement UCMSCs to fat transplantation,then harvested the transplantation tissue at 1week,1 month,2 months after surgery, the weight of the drafted tissue was measured.HE staining was used to analysis the absorption,cysts,fibrosis within the tissue,real -time PCR was performed to detect PPAR -γ,VEGF gene expression.UCMSCs could help to increase the graft tissue weigtht and VEGF to a certain degree,though there was no statistical difference at 1week,1 month and.2 months post -transplantation.UCMSCs can not to increase vascularization and adipogenesis or decrease the absorption of the graft fat significantly,it’s still doubtful to apply UCMSCs in adipose tissue transplantation.
