摘要
为了研究陇东绒山羊生长激素(growth hormone,GH)基因的遗传多态性,确定其等位基因数以及各等位基因间的遗传关系。以陇东绒山羊为研究对象,选择陇东绒山羊GH基因Exon1和Exon4作为候选基因,采用PCR-SSCP分子标记技术和DNA测序技术进行多态性研究。结果表明:在Exon4位点1 450bp处发生了C→T的单碱基突变,产生了产G、H两个等位基因和GG、GH、HH 3种基因型,G基因为优势基因,GG为优势基因型,这为后期开展寻找影响陇东绒山羊经济性状的遗传标记奠定了理论基础。
The aim of this study was to research genetic polymorphisms of growth hormone(GH)gene,confirm the number and genetic relationship of allelic.Exon1 and exon4of Long-dong cashmere goats were studied by PCR-SSCP assay and DNA sequencing.The results showed that a single-base mutant of C→T change at the loci of 1450 bp in exon 4was observed.Two alleles(G and H)and three genotypes(GG,GH and HH)were identified,G was an advantageous gene and GG was an advantageous genotype.The results of this paper were a basis for finding genetic marker which effect on economic characters of long-dong cashmere goats.
