白花蛇舌草高效液相色谱-二极管阵列检测指纹图谱的建立

摘要 目的:建立白花蛇舌草药材的HPLC指纹图谱。方法:采用高效液相色谱-二极管阵列检测法,选用Diamonsil C18(250 mm×4.6 mm,4μm)色谱柱,Easy GuardⅡC18为保护柱,甲醇-0.1%冰醋酸溶液为流动相梯度洗脱,流速1.0 ml·min-1,检测波长254 nm,柱温40℃。采用中国药典委员会颁布的《中药色谱指纹图谱相似度评价系统(2012.130723)》评价不同产地白花蛇舌草图谱的相似性。结果:采用10批白花蛇舌草药材建立HPLC指纹图谱,方法学符合指纹图谱技术要求,10批白花蛇舌草的相似度在0.766-0.985之间。结论:该指纹图谱方法稳定、准确、可靠,为白花蛇舌草药材质量标准的制定提供了科学依据。 OBJECTIVE To establish HPLC fingerprints of Hedyotis diffusa. METHODS High performance liquid chromatography-diode array detection( HPLC-DAD) analysis was performed on Diamonsil C18column(250 mm × 4. 6 mm,4 μm) with Easy GuardⅡ C18 as guard column,methanol and 0. 1% acetic acid as mobile phase in a gradient elution at a flow rate of 1. 0 ml·min^-1,column temperature at 40 ℃. Similarity evaluation software(2012. 130723) issued by Chinese Pharmacopoeia Commission was used to analyze similarity of these ten prints. RESULTS Fingerprints of 10 batches of Hedyotis diffusa were established. Method validation results met requirements of fingerprints. Similarity factors of Hedyotis diffusa from different habitats ranged from 0. 766 to 0. 985. CONCLUSION This method is stable,accurate and reliable,which can provide a scientific basis for quality assessment of Hedyotis diffusa.
