四君子丸含药血清对人肺腺癌SPCA1细胞凋亡及Caspase-3/8/9表达的影响 被引量:8

Effects of Sijunzi decoction containing serum on apoptosis and Caspase-3/8/9 mRNA expression of human lung adenocarcinoma SPCA1 cells

摘要 目的探讨四君子丸含药血清对人肺腺癌SPCA1细胞凋亡及Caspase3/8/9 mRNA表达的影响。方法 30只SPF级雄性大鼠随机分为对照组(n=20)、四君子丸组(n=10)。对照组每日给予1ml/100g生理盐水灌胃,四君子丸组按照每日0.648g/100g灌胃,每天1次,共7d,第7天灌胃给药1.5h后腹主动脉取血分离出血清;采用含10%对照组血清以及10%四君子丸组血清的DMEM培养基分别培养SPCA1细胞;加药处理48h后采用流式细胞仪检测分析SPCA1细胞凋亡率的改变;同时采用Real-time PCR的方法检测SPCA1细胞中Caspase-3/8/9 mRNA表达水平的改变。结果与10%对照血清组相比较,10%四君子丸血清组SPCA1细胞的凋亡率显著增加[(2.33±0.31)%vs(6.13±0.51)%(P<0.01)],细胞中Caspase-3/8/9 mRNA含量均显著上调(P<0.01)。结论四君子丸含药血清可以促进人肺腺癌SPCA1细胞的凋亡,促凋亡可能是四君子丸治疗肺癌的潜在机制。 Objective To investigate the effects of Sijunzi decoction containing serum on apoptosis and Caspase- 3 /8 /9 mRNA expression of human lung adenocarcinoma SPCA1 cells. Methods 30 SPF male rats were randomly divided into control group( n= 20) and the Sijunzi decoction group( n = 10). Rats in control group were given daily 1ml /100 g physiological saline and rats in Sijunzi decoction group were given daily 0. 648 g /100 g Sijunzi pills,once everyday for a week. In seventh days we collected serum from the abdominal aorta 1. 5h after drug treatment. SPCA1 cells were cultured in DMEM medium containing 10% control group serum and 10% Sijunzi decoction group serum respectively. The apoptosis rate and Caspase- 3 /8 /9 mRNA expression level were examined by FCM and Real- time PCR 48 hours after drug treatment. Results The apoptosis ratio of 10% Sijunzi decoction group was significantly increased[( 2. 33 ± 0. 31) % vs( 6. 13 ± 0. 51) %( P〈0. 01) ]compared with 10% control group. and Caspase- 3 /8 /9 mRNA level in SPCA1 cells were all significantly increased( P〈0. 01). Conclusion Sijunzi decoction can promote apoptosis of human lung adenocarcinoma SPCA1 cells,and apoptosis may be a potential mechanism of Sijunzi decoction in the treatment of lung cancer.
