摘要
目的建立香茶菜总三萜及齐墩果酸和熊果酸的含量测定方法,为其质量评价提供依据。方法采用紫外-可见分光光度法,以熊果酸为对照品,5%香草醛冰醋酸-高氯酸为显色剂,于547 nm测定香茶菜中总三萜的含量;采用高效液相色谱法,色谱柱为Welch XB-C18色谱柱(4.6 mm×250 mm,5μm),流动相为甲醇-0.1%甲酸水溶液(85:15),流速为0.6 ml·min-1,柱温为30℃,检测波长为210 nm,测定香茶菜中齐墩果酸和熊果酸的含量。结果不同产地、批次和药用部位的香茶菜总三萜含量为0.13%-1.14%;齐墩果酸和熊果酸在不同的药用部位含量差异显著:根中未检测到,全草中含量分别为0.0128%、0.0388%,地上部位中含量为0.0230%-0.0514%、0.0756%-0.1891%。结论该方法简便、准确,重复性好,可用于香茶菜的质量控制;明确和规范药用部位是其质量控制的前提。
Objective To establish the method for determining the contents of total triterpenoids,oleanolic acid and ursolic acid in Isodon amethystoides( Benth.) C. Y. Wu et Hsuan of different growing places,batches and medicinal parts,and to provide a basic evidence for its quality control. Methods The contents of total triterpenoids were determined by spectrophotometry using ursolic acid as reference substance,and sample solution was colored with 5% vanillin glacial acetic- perchloric acid and the absorbance was measured at 547 nm. The contents of oleanolic acid and ursolic acid were determined by HPLC. Analysis was performed on a Welch XB- C18column( 4. 6 mm × 250 mm,5 μm) with a mobile phase of methanol- 0. 1% formic acid( 85: 15). The flow rate was 0. 6 ml·min^- 1and the column temperature was at 30 ℃. The detection wavelength was at 210 nm. Results The contents of total triterpenoids were ranged from 0. 13% to 1. 14% in the eight samples collected from different growing places,batches and medicinal parts. The contents of oleanolic acid and ursolic acid were 0. 0128%,0. 0388% in herb and 0. 0230 - 0.0514% 0. 0756 - 0. 1891% in aerial parts,respectively,while they are not detected in the root sample. Conclusion The established methods are suitable for the determination of total triterpenoids,oleanolic acid and ursolic acid in Isodon amethystoides,and can be used for the quality control of Isodon amethystoides( Benth.) C. Y. Wu et Hsuan.
