云南万寿菊叶斑病病原菌的鉴定与ITS序列分析 被引量:3

摘要 为明确云南万寿菊叶斑病病原菌种属分类地位,对采自云南蒙自的万寿菊叶斑病病叶,依据科赫法则进行分离、纯化、回接和再分离,证明获得的分离物是叶斑病的致病菌。对其形态特征和培养性状进行观察,并对核糖体DNA-ITS序列进行分析。结果表明,致病菌株在PDA培养基上呈灰褐色;分生孢子梗直立或弯曲,有隔膜;分生孢子倒棒形或卵形,有纵、横隔膜,无喙或具短喙。核糖体DNA-ITS序列与GenBank数据库芸苔生链格孢同源性高达100%。将云南万寿菊叶斑病的病原菌鉴定为芸苔生链格孢Alternaria brassicicola。 The pathogens of marigold leaf spot are different at different regions.In order to determine the pathogen of marigold leaf spot in Yunnan Province,Infected leaves were collected from Mengzi,Yunnan Province.The pathogen was isolated,back-inoculated and re-isolated by Koch's postulates.According to morphological characteristics,culture shapes and the sequence of ribosmal DNA-ITS.The results showed that the fungus colonies on PDA are grayish-brown,conidiophore erect or ascending,septate,conidia obclavate,obovoid,transverse septa,longitudinal septa,pseudorostra have or not.Ribosomal DNA-ITS region sequence analysis indicated that the fungus pathogen and Alternaria brassicicolaisolate had as high as 100% homology.The pathogen was identified as Alternaria brassicicola.
