DHA对人鼻咽癌CNE2细胞增殖的抑制作用及机制研究

摘要 目的研究DHA对鼻咽癌细胞增殖、凋亡的影响。方法CCK8法检测DHA对CNE2细胞增殖的影响;AnnexinV/PI双染流式细胞仪检测DHA对CNE2细胞凋亡的影响:RT—PCR检测DHA处理后CNE2细胞凋亡相关蛋白表达变化情况。结果DHA较对照组明显抑制CNE2细胞增殖。经DHA处理后CNE2细胞中Bcl-2、Bcl—xl表达下降,且呈剂量依赖性。结论DHA可有效抑制Bcl-2、Bcl—xl的表达而发挥抑制癌细胞增殖、诱导凋亡效应。 Objective To study the influence of DHA on the proliferation and apoptosis of CNE2 cells in nasopharyngeal carcinoma. Methods CCK8 assay was used to investigate the influence of DHA on the proliferation of CNE2 cells. Flow eytometry (FCM) were used to examine the effect of DHA on the apoptosis of CNE2 cells. Reverse transcription polymerase chain reaction (RT-PCR) was applied to detect the mRNA expressions of Bcl-2 and Bcl-xl in CNE2 cells. Results DHA more significantly inhibited the proliferation of CNE2 cells than the control. FCM method indicated that the apoptosis of CNE2 cells was significantly higher after exposure to DHA than to the control (P 〈 0.05). The mRNA levels of Bcl-2 and Bclxl were significantly decreased in the DHA group. Conclusions DHA can inhibit the proliferation and induce the apoptosis of CNE2 cells down-regulating the mRNA expressions of Bcl-2 and Bcl-xl.
