电致化学发光技术用于单细胞表面磷脂酰胆碱的分析

摘要 文章利用电致化学发光技术分析单细胞膜表面磷脂酰胆碱。在电极表面培养细胞,利用特定的磷脂酶D与细胞表面磷脂酰胆碱反应生成胆碱,然后胆碱再与胆碱氧化酶反应产生过氧化氢;在正电位下,鲁米诺作为发光试剂存在时诱导过氧化氢发光,通过对光强的检测,实现对单细胞表面磷脂酰胆碱的检测。 Here, phosphatidylcholines (PCs) in plasma membrane at single cells was analyzed using luminol electrochemiluminescence. The detection process involved that the cleavage of PCs at cellular membrane by PC-specific PLD to generate choline, and the following reaction of choline with choline oxidase for the production of hydrogen peroxide. In presence of luminol, hydrogen peroxide induced luminescence under the positive potential so that PCs at cellular membrane were determined.
