摘要 建立了高效液相色谱-蒸发光散射检测器(HPLC-ELSD)测定低聚木糖中的木糖和木二糖含量的方法。以Prevail TM Carbohydrate ES糖柱(250×4.6,5μm)为色谱柱,乙腈—水(72∶28,V/V)为流动相,流速为1.0 m L/min,柱温为25℃,进样量为10μL,ELSD漂移管温度为80℃,载气流速为2.0 L/min。结果表明,木糖和木二糖分别在0.08~1.00 mg/m L和0.15~2.00 mg/m L质量浓度范围内线性关系良好,检出限分别为0.01mg/m L和0.02 mg/m L,定量限分别为0.03 mg/m L和0.05 mg/m L,样品加标回收率为97%~102%。 An analytical method was established for the determination of xylose and xylobiose by HPLC-ELSD. The optimal conditions were as follows: column was PrevailTM Carbohydrate ES (250×4.6, 5 μm), mobile phase: mixture of acetonitrile and water (72 : 28), flow rate: 1.0mL/min, column temperature: 25 ℃, injection volume: 10 μL, drift tube temperature: 80 ℃, flow rate of carrier gas: 2.0 L/min. Under the optimal conditions, the calibration curve was linear over the content of xylose and xylobiose in the range of 0.08-1.00 mg/mL and 0.15-2.00 mg/mL, the detection limits (LOD) were 0.01 mg/mL and 0.02 mg/mL, the quantification limits (LOQ) were 0.03 mg/mL and 0.05 mg/mL, respectively. The spiked recoveries of xytose and xylobiose were 97 %-102 %.
