摘要
目的探讨白藜芦醇对卵巢上皮细胞癌侵袭能力的影响。方法人卵巢癌细胞分为三组,低转移组,高转移组,高转移+白藜芦醇组,分别提取蛋白和RNA,Western Blot测定MMP-7、Sirt-1、AP-1的蛋白表达水平,RT-PCR测定MMP-7的基因转录水平。结果白藜芦醇可以明显的激活高转移卵巢上皮细胞癌当中Sirt-1的作用,与高转移组相比,Sirt-1的表达明显增加(P<0.05),白藜芦醇抑制卵巢上皮细胞癌当中AP-1和MMP-7的表达(P<0.05)。结论白藜芦醇具有明显的降低卵巢上皮细胞癌侵袭性的药理作用,这种作用和其上调Sirt-1的表达密切相关。
Objective To study the effects of resveratrol in ovarian epithelial carcinoma and explore the molecular mechanism. Methods Human ovarian cancer cells was divided into three groups, (A) low metastasis group (B) high metastasis group (C) high metastasis group with resveratrol, then extraction protein and RNA respectively,Western blot analysis were used to determine the effect of resveratrol on the expression levels of SIRT1, AP-1, and MMP-7 proteins and RT- PCR assay were used to determine the genetic transcription level of MMP-7. Results Western Blot analysis and RT-PCR revealed that the levels of MMP-7 and AP-1 were significantly upregulated in high metastasis ovarian epithelial carcinoma, however resveratrol attenuated the levels of MMP-7 and AP-1 (P 〈 0.05), correspondingly, resveratrol could increase the expression of Sirt 1(P 〈 0.05). Conclusion Resveratrol could inhibit metastasis of ovarian epithelial carcinoma mediated by Sirtl activation.
