益肾化浊法对慢性脑缺血血管性痴呆大鼠脑组织海马HIF-1α和VEGF表达的影响 被引量:7

Influence of Yishen Huazhuo Therapy on expression of HIF-1α and VEGF in hippocampus of rats with chronic ischemic vascular dementia

摘要 目的观察益肾化浊法对慢性脑缺血血管性痴呆大鼠脑组织海马低氧诱导因子-1α(hypoxia-inducible factor-1,HIF-1α)和血管内皮生长因子(vascular endothelial growth factor,VEGF)表达的影响。方法采用双侧颈总动脉永久性阻断法建立血管性痴呆大鼠模型,造模后7天随机分为假手术组,模型组和聪圣颗粒低、中、高剂量组和尼麦角林片组,给药治疗60天后,采用Western-blot及Real Time-PCR方法检测大鼠海马HIF-1α、VEGF表达水平。结果与假手术组比较,模型组大鼠HIF-1α和VEGF的蛋白表达量和mRNA的表达量均升高(P<0.05);与模型组比较,聪圣颗粒中、高剂量组可升高模型组HIF-1α及VEGF的蛋白和mRNA的表达(P<0.05,P<0.01)。结论益肾化浊法对慢性脑缺血血管性痴呆大鼠作用机制可能与调控海马区HIF-1α和VEGF的表达有关。 Objective To observe the influence of Yishen Huazhuo Therapy on expression of hypoxia-inducible factor-1( HIF-1α) and vascular endothelial growth factor( VEGF) in hippocampus of rats with chronic ischemic vascular dementia. Methods Vascular dementia rat models were established by using bilateral carotid artery block method. 7 days after animal model establishment,rats were randomly divided into sham-operated group,model group,high,middle and low dosage of Shengcong Particle groups and nicergoline tablets group. After 60 days of treatment,the expression level of HIF-1α and VEGF in hippocampus of rats was tested by using Western blot and Real time PCR. Results Compared with sham-operated group,the protein and mRNA expression of HIF-1α and VEGF in hippocampus of rats in model group was increased significantly( P〈0. 05). Compared with model group,the protein and mRNA expression of HIF-1α and VEGF in hippocampus of rats in high and middle dosage of Shengcong Particle group was increased significantly( P〈0. 05,P〈0. 01). Conclusion Yishen Huazhuo therapy could improve the status of rats with chronic ischemic vascular dementia and the mechanism might be related with the regulation on expression of HIF-1α and VEGF in hippocampus of rats.
