摘要
目的:研究氯化铝对体外培养大鼠睾丸支持细胞的雄激素结合蛋白(ABP)mRNA表达的影响。方法:体外培养大鼠睾丸支持细胞为模型,分为对照组、低剂量组、中剂量组、高剂量组(0,300,400,500μmol/L)氯化铝染毒睾丸支持细胞,培养24h后采取MTT法检测细胞活性,RT-PCR法检测ABP mRNA的表达水平。结果:随着氯化铝染毒剂量的增大,ABP mRNA的表达下降。高剂量染毒组与对照组相比较,ABP mRNA的表达差异具有统计学意义(P<0.05)。结论:氯化铝对体外培养的睾丸支持细胞具有毒性作用,抑制ABP mRNA的表达,从而损伤支持细胞功能,导致生殖功能障碍。
Objective: To explore the effects of aluminum chloride on expression of androgen binding protein mRNA in Sertoli cells of rat. Methods: Rat Sertoli cells were cultured, purified and iden- tified and treated with different concentrations (0,300,400,500 μmol/L) of aluminum chloride for 24 h, cell viability was evaluated by MTT method. The levels of ABP mRNA expression were measured by RT-PCR. Results: The androgen binding protein mRNA levels decreased with the increase of Aluminum chloride concentration in Sertoli cell cultures. Compared with control group, the difference of ABP mRNA expression of was statistically significance (P〈0. 05). Conclusion: Aluminum chloride is a reproductive toxicant which disrupts the transcription of ABP in rat Sertoli cells so as to result in reproductive dysfunction.
