双氢青蒿素对人胶质瘤U87细胞迁移和侵袭的影响 被引量:2

摘要 目的探讨双氢青蒿素(Dihydroartemisinin,DHA)对胶质瘤U87细胞迁移和侵袭能力的影响。方法采用CCK8法检测胶质瘤U87细胞生存率;划痕实验和transwell侵袭试验检测DHA对胶质瘤U87细胞迁移和侵袭能力的影响;Western blotting分析DHA对细胞基质金属蛋白酶(MMP-2、MMP-9)蛋白表达的改变。结果 CCK8显示DHA对胶质瘤U87细胞的增殖抑制呈浓度和时间依赖性(P<0.05);与对照组比较,经DHA处理后胶质瘤U87细胞迁移和侵袭能力明显降低,相对应的MMP-2、MMP-9蛋白表达下降(P<0.05)。结论 DHA可有效抑制胶质瘤U87细胞迁移和侵袭能力,其机制可能与DHA降低细胞MMP-2、MMP-9的蛋白表达相关。 Objective To investigate the effects of dihydroartemisinin (DHA) on migration and invasion in U87 glioma cells. Methods The U87 cells were cultured with different concentrations of DHA, and CCK8 assay was performed to evaluate the cell survival. Cell migration and invasive capacities were evaluated by means of scratch wound assay and transwell matrix penetration assay. The activity and expressions of invasion-related proteins matrix-metallopeptidase 2 ( MMP-2 ) and MMP-9 were analyzed by Western blotting . Results DHA caused concentration-dependent and time-dependent inhibition of the proliferation of U87 cells (P〈0.05). Cells treated with DHA showed significantly decreased U87 cells migration and invasion capabilities. Western blotting demonstrated that the protein expressions of MMP-2 and MMP-9 were suppressed by DHA in U87 cells (P〈0.05). Conclusions DHA may play its role inhibiting glioma cell line U87 migration and invasion via suppressing the expressions of MMP-2 and MMP-9.
