摘要
目的探讨胶质瘤中波形蛋白、NF-κB表达相关性及其与肿瘤侵袭性的关系。方法采用免疫组织化学染色检测波形蛋白、NF-κB和MMP-7在28例低级别(Ⅰ、Ⅱ)胶质瘤和18例高级别(Ⅲ、Ⅳ)胶质瘤中的表达,采用RT-PCR技术检测46例胶质瘤波形蛋白基因表达情况。结果高级别胶质瘤波形蛋白、NF-κB和MMP-7表达均高于低级别胶质瘤(χ2分别为4.44、5.22、6.47,均P<0.05)。波形蛋白表达与NF-κB表达呈正相关(r=0.414,P<0.05)。结论波形蛋白与NF-κB在恶性胶质瘤均高表达,且两者呈正相关,协同MMP-7高表达而促进肿瘤细胞迁徙及侵袭。
Objective To investigate the expressions of vimentin and NF-κB in invasive glioma and relationship between the expressions of both vimentin and NF-κB and the invasion of glioma. Methods Immunohistochemical staining was used to detect the differential expressions of vimentin, NF-κB and MMP-7 in 28 low-grade (gradesⅠandⅡ) gliomas and 18 high-grade (gradesⅢandⅣ) gliomas. The gene expression of vimentin was detected by RT-PCR in 46 gliomas. Results Vimentin, NF-κB and MMP-7 were all more highly expressed in high-grade gliomas than the low-grade those (χ2=4.44, 5.22, 6.47, all P〈0.05). The expressions of vimentin and NF-κB was of positive correlation (r =0.414, P〈0.05). Conclusions The high expression of NF-κB promotes the expression of vimentin, which collaborates with MMP-7, leading to the invasion and migration of glioma.
