补肾益气方对反复自然流产患者外周血调节性T细胞的影响

Effect of Bushen Yiqi formula on regulatory T cells in patients suffered from recurrent spontaneous abortion

目的探讨补肾益气方对反复自然流产(recurrent spontaneous abortion,RSA)患者外周血调节性T细胞的影响。方法随机选取15例健康女性(对照组)和30例RSA患者,RSA患者诊断受孕后口服中药补肾益气方,2周后流式细胞仪检测RSA患者服药前后及健康对照组调节性T细胞数量,ELISA法检测外周血肿瘤坏死因子α(tumor necrosis factor-α,TNFα)、干扰素γ(interferon-γ,IFNγ)及白细胞介素-10(interleukin-10,IL-10)水平变化。结果中药治疗2周后RSA患者外周血调节性T细胞数量上升,IL-10水平上升,均明显高于服药前及健康对照组(P<0.05),Th1类细胞因子TNFα和IFNγ水平明显下降(P<0.05)。结论补肾益气方能够上调RSA患者血调节性T细胞水平,改变母体细胞因子分泌状态,诱导母胎免疫耐受,保护胎儿不被免疫细胞排斥。

Objective To investigate the effect of Bushen Yiqi formula on regulatory T cells in patients suffered from recurrent spontaneous abortion（RSA）.Methods Fifteen healthy control women and 30 patients suffered from RSA were selected randomly.Patients were administered with herbs as soon as the pregnancy was established.The blood of RSA patients was collected two weeks later after herbs treatment.The frequency of regulatory T cells in blood was examined by flow cytometry and the expression of tumor necrosis factor-α （TNFα）,interferon-γ （IFNγ） and interleukin-10 （IL-10） was determined by ELISA.Results The number of regulatory T cells was elevated in patients with herbs compared with healthy controls and themselves before treatment（P＜0.05）.IL-10 expressed highly in patients with herbs,while lowly in patients before herbs treatment and healthy controls（P＜0.05）.Conversely,the level of TNFα and IFNγ in patients with herbs was lower than themselves before treatment and healthy controls.Conclusions Bushen Yiqi formula protects the fetus from maternal immune cell attack by upregulating the regulatory T cells number and balancing materral cytokine expression,and inducing immune tolerance for both mom and infant.
