摘要 目的:建立HPLC法同时测定肾炎四味胶囊中黄芩苷、黄芩素、汉黄芩苷、汉黄芩素等4种成分。方法:采用Agilent ZORBAX SB—C18(250mm×4.6mm,5μm)色谱柱,流动相为甲醇-0.4%磷酸溶液,梯度洗脱,流速为1.0ml·min^-1,检测波长为278nm,柱温为30℃,进样量为10μl。结果:上述色谱条件下,4种成分分离较好,黄芩苷、黄芩素、汉黄芩苷和汉黄芩素线性关系良好,线性范围分别为0.060~1.203μg(r=0.9999)、0.026~0.521μg(r=0.9999)、0.031—0.622μg(r=0.9998)、0.025—0.497μg(r=0.9996),加样回收率分别为97.9%(RSD=1.72%)、101.8%(RSD=1.86%)、102.1%(RSD=1.35%)、99.3%(RSD=1.12%)(n=9)。结论:本方法准确度高、重复性好,能同时测定肾炎四味胶囊中黄芩苷、黄芩素、汉黄芩苷、汉黄芩素等4种成分的含量。 Objective: To establish an HPLC method for the simultaneous determination of four kinds of flavonoids ( baiealin, wogonoside, baiealein and wogonin) in Shenyansiwei ,capsules. Methods: An Agilent ZORBAX SB-C18 eolumn(250mm×4.6mm,5μm) was used, the mobile phase consisted of methanol-0.4% phosphoric acid solution with gradient elution, the flow rate was 1.0 ml ·min^-1 the detection wavelength was set at 278 nm, the column temperature was 30℃, and the injection volume was 10 μl Results: The four kinds of flavonoids were well separated and showed good linearity. The linear range of baiealin, wogonoside, baiealein and wogonin was O. 060-1. 203 μg( r = 0 999 9 ) ,0. 026-0. 521 μg( r = 0. 999 9 ) ,0. 031-0. 622 μg( r = 0. 999 8 ) and 0. 025-0. 497 μg( r = 0.999 6 ), respectively. The average reeovery of the four kinds of flavonoids was 97.9% ( RSD = 1.72%, n = 9 ), 101.8% ( RSD = 1.86%, n = 9), 102.1% (RSD = 1.35%, n = 9) and 99.3% (RSD = 1.12%, n = 9 ), respectively. Conclusion: The method is rapid, simple, reliable and aeeurate, which can be successfully used in the quantification of the four constituents in Shenyansiwei capsules.
