可溶性血管内皮钙粘素与脑胶质瘤患者生存期相关性研究 被引量:3

Study on the correlation of soluble vascular endothelial cadherin and survival of patients with glioma

摘要 目的 分析结构改变的血管内皮(vascular endothelial,VE)-钙粘素(包括酪氨酸磷酸化和裂解)在脑胶质瘤患者中的表达及临床意义.方法 研究对象为60例在本院手术的脑胶质瘤患者,采用免疫组化染色和Western blot观察肿瘤标本的VE-钙粘素的表达及磷酸化情况.通过去糖基化分析和酶联免疫吸附测定(enzyme-linked immunosorbent assay,ELISA)定量分析脑胶质瘤患者血清中裂解VE-钙粘素浓度(soluble VE-cadherin,sVE),分析其与患者生存期的相关性.结果 相对于非肿瘤组织,VE-钙粘素在人胶质瘤组织中高表达(P<0.05),VE-钙粘素酪氨酸磷酸化表达也明显增高(P<0.05).与此同时,胶质瘤患者血清中sVE浓度增高,患者(sVE<840 ng/ml)中位进展期为1 095 d,患者(840<sVE<1 247 ng/ml)中位进展期为365 d,患者(sVE>1 247 ng/ml)中位进展期为204 d,同组织学分级无关.结论 sVE的表达与患者的预后不良有关,可以作为脑肿瘤预后标志物。 Objective To analyze the structural modifications of vascular endothelial-cadherin expression and clinical significance in human glioma.Methods 60 surgery patients with brain gliomas were screened.The expression of vascular endothelial cadherin and phosphorylation status were tested by immunohistochemical and western blotting.Deglycosylation and ELISA were used to analyze serum soluble vascular endothelial cadherin (sVE) in patients with glioma.Finally,Cox model was used to investigate sVE correlation with the survival of patients.Result The high expression of vascular endothelial cadherin in human glioma tissues,tyrosine phosphorylation of vascular endothelial cadherin was showed compared with normal tissue(P〈0.05).The serum sVE level increased in patients with glioma,and patients with sVE〈840 ng/ml had amedian time to progression of 1095 days.Patients with 840〈sVE 〈1247 ng/ml had a median time to progression of 365 days whereas patients with sVE 〉 1247 ng/ml had a median time to progression of 204 days,irrespective to histopathological grade of tumors.Conclusion The results suggest that sVE should be examined as candidate biomarkers of brain tumor,with promising applications in oncology.
