Role of lysosomal-associated membrane protein 3 in the tumorigenesis of colon cancer

摘要 目的 观察溶酶体相关膜蛋白3(LAMP3)在结肠癌中的作用.方法 应用免疫组织化学法检测126例结肠癌临床样本中LAMP3的表达,Kendall' tau-b法进行相关分析;Kaplan-Meier 法绘制生存曲线,Log-rank法对生存分析进行检验;Cox比例风险回归模型分析独立预后影响因素.结果 LAMP3在癌旁组织中呈低表达,在结肠癌组织中表达不一,有些患者结肠癌组织中LAMP3低表达,与正常组织类似,有些呈高表达;LAMP3的表达与与患者性别、年龄、肿瘤大小、分化程度、淋巴转移、远处转移、复发等因素之间无明显相关,与肿瘤TNM分期之间明显相关(P<0.01).Cox比例风险回归模型分析显示LAMP3表达[风险比(HR)=2.009,95%可信区间(CI):1.602 ~5.890,P<0.05]是结肠癌患者生存的独立影响因素;Kaplan-Meier生存分析表明,与LAMP3低表达的患者比较,LAMP3高表达的结肠癌患者预后较差(P<0.05).结论 LAMP3是结肠癌患者预后的独立影响因素. Objective To investigate the role of lysosomal-associated membrane protein 3 (LAMP3) expression in the tumorigenesis of colon cancer.Methods Expression of LAMP3 in colon cancer tissues and normal colon tissues were detected by immunohistochemistry;Correlation between variables was analyzed by Kendall' s tau-b method.Survival curves were plotted using Kaplan-Meier method with Log-rank test.Cox proportional hazard model was used to evaluate the independent prognostic factors.Results LAMP3 expression was low in normal colon tissues,while high in some colon cancer tissues.LAMP3 expression level did not correlated with gender,age,tumor size,differentiation,lymph nodal metastasis,distant metastasis and recurrence.LAMP3 expression statistically significantly correlated with TNM stages (P < 0.01).Cox regression analysis indicated that LAMP3 expression was independent prognostic factors of colon cancer patients [hazard ratio (HR) =2.009,95% confidence interval (CI):1.602-5.890,P < 0.05].Kaplan-Meier survival analysis showed that,compared with patients with low LAMP3 expression,patients with high LAMP3 expression had poorer overall survival (P < 0.05).Conclusion LAMP3 is an independent prognostic factor of colon cancer patients.
