寻常性银屑病患者外周血干扰素γ和肿瘤坏死因子α及其受体mRNA的测定 被引量:3

Expression of IFN-γ TNF-α and their receptor mRNA in patients with psoriasis vulgaris

摘要 目的研究外周血干扰素(IFN)-γ,mRNA和肿瘤坏死因子(TNF)-α及其受体mRNA在寻常性银屑病患者中的表达。方法选择2010年8月至2013年8月本院50例寻常性银屑病患者为观察组,将观察组患者按是否为进行期分两亚组,进行期为观察组A(28例),静止期为观察组B(22例);选另同期本院体检中心体检的健康者30例为健康对照组。采集各组外周静脉血,进行血清IFN-γ和TNF-α水平检测,同时检测其PBMC中IFN-γ受体mRNA和TNF—α受体mRNA的表达情况。结果观察组患者血清IFN-γ和TNF-α水平均高于健康对照组[(8.46±2.72)比(5.61±2.43)pg/mL;(44.28±8.61)比(3.26±6.85)pg/mL,均P〈0.05];观察组A中IFN-γ受体mRNA及TNF-α受体mRNA的表达水平高于观察组B[(1.21±0.23)比(0.75±0.38);(2.63±2.18)比(1.58±1.17),均P〈0.05],而观察组B中IFN-γ受体mRNA及TNF—α受体mRNA的表达水平高于对照组,但差异无统计学意义[(0.75±0.38)比(0.64±0.27);(1.58±1.17)比(1.24±1.23),均P〈0.05];PASI评分不因IFN-γ受体mRNA和TNF—α受体mRNA表达水平的变化而变化(r=0.15,P〉0.05)。结论银屑病患者进行期时,IFN-γ受体mRNA及TNF-α受体mRNA的表达明显升高,银屑病的发病作用机制与IFN-γ受体mRNA及TNF-α受体mRNA的表达可能相关。 Objective To study the expression of interferon-γ(IFN-α) ,tumor necrosis factor-α (TNF-α) and their receptor mRNA in patients with psoriasis vulgaris. Methods 50 patients with psoriasis vulgaris treated from August 2010 to August 2013 in our hospital were selected as observation group. The observation group were also divided into group A(28 patients in the active stage) and group B(22 patients in the stationary phase). In addition ,30 healthy subjects were selected as control group. The peripheral venous blood of the two groups was collected. Levels of IFN-γ and TNF-α were measured. The expression of PBMC IFN-γ and TNF-α receptor mRNA was detected. Results IFN-γ and TNF-α levels of observation group were significantly higher than that of control group [ (8.46 ± 2.72) vs ( 5.61 ± 2.43 ) pg/mL; (44.28 ± 8.61 ) vs ( 3.26 ± 6.85 ) pg/mL, both P 〈 0.05 ]. The expression of IFN-γ and TNF-α receptor mRNA of group A was significantly higher than that of group B[ (1.21 ±0.23)vs(0.75 ±0.38) ;(2.63 ±2.18)vs(1.58 ±1.17) ,both P 〈0.05]. The expression of IFN-γ and TNF-α receptor mRNA of group B was significantly higher than that of control group [ (0.75±0.38)vs(0.64 ±0.27);(1.58 ±1.17)vs(1.24 ±1.23),both P〈0.05]. PASI scores were changed with the different expression levels of IFN-γ and TNF-α receptor mRNA ( r = 0. 15, P 〉 0. 05 ). Conclusion In the active stage of psoriasis, the expressions of IFN-γ and TNF-α receptor mRNA were significantly increased. The psoriasis pathogenesis mechanism is correlated with the expressions of IFN-γ and TNF-α receptor mRNA.
