摘要
[目的]建立盐生肉苁蓉药材中2种水溶性成分尿囊素和半乳糖醇的薄层色谱分析方法。[方法]采用硅胶G薄层板,分别对乙酸乙酯∶丙酮∶甲酸、氯仿∶异丙醇∶乙酸等6种不同的展开剂及25%对二氨基苯甲醛、10%硫酸乙醇、3%酸性高锰酸钾溶液、10%香草醛浓硫酸4种不同的显色剂进行考察,确定盐生肉苁蓉提取物中尿囊素、半乳糖醇的最佳薄层分析条件,并对不同产地盐生肉苁蓉中的2种成分进行鉴定。[结果]尿囊素的展开剂为乙酸乙酯∶丙酮∶甲酸(8∶1∶3),显色剂为25%对二氨基苯甲醛溶液;半乳糖醇的展开剂为乙酸乙酯∶异丙醇∶水∶甲酸(3∶4∶2∶1),显色剂为3%酸性高锰酸钾溶液。不同产地盐生肉苁蓉中均含有尿囊素和半乳糖醇。[结论]该方法快速简便、专属性强、重现性好,可快速鉴别肉苁蓉药材,是全面控制和评价盐生肉苁蓉药材质量的重要补充。
[Objective] To establish TLC analysis method of 2 kinds of water-soluble ingredients Allantoin and Dulcitol in Cistanche salsa.[Method]Screening TLC conditions with 6 kinds of developing solvent system and 4 kinds of chromogenic reagents. In addition,TLC results of four authentic samples from different producing area were compared. [Result] TLC conditions of Allantoin was Ethyl acetate∶ Acetone∶ Formic acid( 8∶1∶3) developing with 25% p-Dimethylaminobenzaldehyde as chromogenic reagent. TLC conditions of Dulcitol was Ethyl acetate∶Isopropanol∶ Water∶ Formic acid( 3 ∶ 4 ∶ 2 ∶ 1) developing with 3% acidic Potassium Permanganate as chromogenic reagent. [Conclusion] The method was quick and easy to operate. The repeatability and specificity were reliable. The method was suitable for the quality control of Cistanche salsa.
