响应面法优化以改良玉米浆为原料产β-胡萝卜素的液体发酵培养基

摘要 玉米浆是玉米粒用亚硫酸浸泡提取玉米淀粉后的副产物,主要成分为玉米可溶性蛋白及其降解物(如肽类、各种氨基酸等),另外还含有乳酸、可溶性糖类等营养成分,可作为发酵工业的原料。由于未处理玉米浆中含有多种金属离子,某些金属离子可能会对微生物的发酵过程产生不利影响,一定程度上限制了其在发酵生产上的应用。本研究采用Na2CO3对玉米浆进行预处理,离心去除沉淀后作为三孢布拉氏霉(Blakeslea trispora)的发酵原料。研究发现当100m L玉米浆中添加1.17g Na2CO3时,β-胡萝卜素产量达到最大,比未处理玉米浆提高20.67%。采用响应面法对三孢布拉氏霉液体发酵培养基进行了优化,经分析得到β-胡萝卜素产量最大时培养基中显著性因素含量为:葡萄糖8.70%、KH2PO4 0.51%,β-胡萝卜素产量可达到84.76mg/L,比优化前提高了29.93%。 Com syrup is byproduct that corn kemels soaked with sulfurous acid after extracting corn starch, the main ingredient is corn soluble protein and degradation products (such as peptides, amino acids, etc.), also contains nutrients with sulfurous acid, soluble sugars, is used as raw material in fermentation industry. But untreated com syrup contains a variety of metal ions, certain metal ions microbial fermentation process may be adversely affected, to some extent, limits its application in the production of fermentation. In this study, Na2CO3 pretreatment of com syrup, and centrifuged to remove precipitated as blakeslea trispora fermentation feedstock. The study found corn syrup per 100mL when adding 1.17g Na2CO3, β-carotene production reached a maximum, than the untreated com syrup 20.67% increase. Response surface methodology was used to optimize the liquid fermentation medium by blakeslea trispora. Optimized to get maximum β -carotene production media by analyzing the content of significant factors : glucose 8.70%, KH2PO4 0.51%, β-carotene production could reach 84.76mg/L, compared to the previous increase of 29.93% optimization.
