工业区与非工业区孕妇及胎儿血浆TNF-α和VEGF的比较

Comparative study on maternal and fetal TNF-α and VEGF in industrial and non-industrial areas

摘要 目的探讨孕期暴露于环境中钴、镍、铜、砷、硒对肿瘤坏死因子-α(TNF-α)、血管内皮生长因子(VEGF)的影响。方法于2011年5—10月,在工业区和非工业区随机选择正常孕妇各30人,采集分娩前母血和分娩时脐带血,测定母血和脐血中钴、镍、铜、砷、硒和TNF-α、VEGF的浓度。结果工业区母血钴、砷、硒浓度高于非工业区(P<0.01),脐血钴、铜、砷、硒浓度高于非工业区(P<0.01,P<0.05)。工业区母血和脐血TNF-α浓度高于非工业区(P<0.05),脐血VEGF浓度低于非工业区(P<0.05)。工业区母血TNF-α与砷、硒呈正相关(P<0.05),脐血TNF-α与钴、铜、硒呈正相关(P<0.05);工业区母血VEGF与钴、砷、硒呈负相关(P<0.01,P<0.05),脐血VEGF与铜呈正相关(P<0.05),与钴、砷、硒呈负相关(P<0.05)。结论孕期暴露于环境中高浓度的钴、镍、铜、砷、硒可影响母体和胎儿TNF-α和VEGF的分泌,导致TNF-α表达增加、VEGF生成减少。 Objective To explore the impact of environmental cobalt (Co),nickle (Ni),copper (Cu),arsenic (As),selenium(Se) exposure on the levels of maternal and fetal tumor necrosis factor-α (TNF-α,vascular endothelial growth factor (VEGF) in plasma. Methods A total of 30 normal pregnant women were randomly selected in industrial and non-industrial areas respectively, then maternal, cord blood were gathered for determination of levels of cobalt, nickle, copper, arsenic, selenium and TNF-α,VEGF. Results The results of elements analysis of maternal and cord blood in two areas showed that the concentrations of cobalt, arsenic and selenium of maternal blood in industrial area were higher than those in non-industrial area (P〈0.O1). The concentrations of cobalt, copper, arsenic, selenium of cord blood in industrial area were higher than those in non- industrial area (P〈0.01 ,P〈0.05) . The results of analysis of TNF-α and VEGF in plasma indicated that the levels of TNF-α of maternal and cord blood in industrial area were both higher than those in non-industrial area (P〈0.05); The level of VEGF of cord blood in industrial area was lower than that in non-industrial area (P〈0.05). In industrial area,positive correlation was observed between TNF-α in maternal blood and arsenic, selenium in maternal blood (P〈O.05), also between TNF-α in cord blood and cobalt, copper, selenium in cord blood (P〈0.05); While negative correlation was found between VEGF in maternal blood and cobalt,arsenic,selenium in maternal blood (P〈0.01,P〈0.05),also between VEGF in cord blood and cobalt, arsenic, selenium in cord blood (P〈0.05),but the correlation between VEGF and copper in cord blood was positive (P〈0.05). Conclusion The TNF-α and VEGF in plasma can be affected by high levels of cobah,nickle,copper,arsenic,selenium during pregnancy,and the exposure to these elements will increase the TNF-α expression, decrease VEGF.
