板栗花黄酮的抗氧化作用及其对Hela细胞活力的影响 被引量:9

摘要 采用水杨酸法、邻苯三酚自氧化法和三价铁离子法分别测定板栗花黄酮清除羟自由基(·OH)和超氧阴离子自由基(O2-·)的能力及其总还原力,以研究板栗花黄酮的抗氧化作用;采用MTT法测定纯化板栗花黄酮对Hela细胞活性的影响。结果表明:板栗花黄酮具有较强的还原力并能有效清除·OH,其总还原力和·OH清除率显著高于VC对照组(p<0.05),可达0.605和94.97%;纯化后板栗花黄酮的总还原力和清除·OH能力较纯化前明显增强。但板栗花黄酮对O2-·的清除率明显低于VC对照组。同时纯化后板栗花黄酮能显著抑制Hela的细胞活力,在实验浓度范围内,抑制率最高至81%,呈剂量-效应性。因此,板栗花黄酮具有较强的抗氧化活性和抑制Hela细胞活力的作用,本文为板栗花的综合利用与开发提供了一定的数据支持。 Antioxidant activity of the flavonoids from chestnut flower was studied by determining free radicals scavenging abilities including superoxide anion free radicals (O2-.) and hydroxyl free radicals (-OH),and total inducing power using the salicylic acid assay,pyrogallol autoxidation assay and ferric reducing antioxidant assay,respectively. And the influence of flavonoids-purified on the Hela cellular viability was also investigated by MTT assays. The results showed that the scavenging -OH ability and total reducing power of flavonoids from chestnut flower were obviously stronger than those of the Vc control,and the fiavonoids-purified sample exhibited the better scavenging .OH ability and total reducing power compared to the flavonoids-cruded sample. The highest scavenging ability on .OH and the strongest total reducing power were 0.605 and 94.97% ,respectively. However,the scavenging ability of before or after fiavonoids-purified samples on O2-. was less effective than that of Vc control. Furthermore,MTT data suggested that the purified flavonoids sample exhibited the powerful cytotoxic activity on Hela cells which appeared to be concentration dependent,and the highest inhibitory cellular viability was 81% within the experimental concentration. Therefore,the flavonoids from chestnut flower had the stronger antioxidant activity and inhibit the growth of Hela cells,and those findings provided the data to support for the comprehensively utilizating and developing chestnut flower.
