摘要
目的:探讨Wnt蛋白(Wnt3a)联合骨形态发生蛋白(BMP7)对骨髓间充质干细胞(BMSCs)成骨分化的作用。方法:选择SD大鼠12只,进行骨髓间充质干细胞的分离、纯化、培养,并采用不同的方法进行骨髓间充质干细胞的体外诱导分化,按干预的不同分为4组:Wnt3a组、BMP-7组、联合诱导组以及对照组,对比不同方法对成骨分化的诱导作用差异。结果:联合诱导组的IA值为(121627±261),显著低于Wnt3a组、BMP-7组、对照组的(147173±348)、(146557±361)、(182659±409),并且其有大量橘红色矿化结节形成,较其他三组更多,组间差异比较均具有统计学意义(P<0.05)。结论:Wnt3a联合BMP7诱导BMSC成骨分化的效果显著优于单独采用Wnt3a或BMP7进行诱导,值得在临床上推广使用。
Objective: To Investigate the differentiation effect of Wnt3a in combination with BMP7 on BMSC osteogenesis. Methods:Select 12 SD rats ,detach ,purify and cultivate BMSCs,externally induct and differentiate BMSCs with different methods, divide them into 4 groups according to the interventions: Wnt3a group,BMP-7 group,associated group and control group ,compare the differences of different methods in in- ducting the differentiation of osteogenesis. Results:The IA value of the associated group was obviously higher than that of the Wnt3a group,BMP-7 group and control group, the figure was (121627!261),(147173~ 348) ,(146557+361),(182659~409)respectively. And a large number of orange mineralized nodules formed in associated group, which was much more than that in the other groups , to compare these groups, the differ- ences were statistically significant(P〈0.05). Conclusion:It is more effective to induct BMSC osteogenesis to differentiate with Wnt3a in combination with BMP7 than with Wnt3a or BMP7 alone,it is worthy of clinical promotion and application.
