摘要 环介导等温扩增(LAMP)技术于2000年由日本学者提出,其反应原理是由两对特异性引物,针对靶基因的6个不同区域进行特异性识别,在DNA聚合酶作用下,恒温条件进行链置换扩增。由于LAMP技术具有高灵敏度、强特异性、快速简便、成本低廉等特点,使得其广泛应用于病原菌的检测、胚胎性别鉴定、食品卫生检验等方面。论文针对LAMP技术的研究背景、反应原理、应用等方面进行了综述,总结出不同因素对LAMP反应的影响,对其优势和局限性进行了讨论,并对LAMP的发展前景进行了展望。 Loop-mediated isothermal amplification(LAMP)was proposed by a Japanese scholar in 2000. Aset of LAMP primers was designed based on the sequence of target, which enabled the identification of six independent regions of the target gene. The reaction was carried out under the constant temperature with the Bst DNA polymerase. It has been widely applied in the pathogen detection, sexing embryos and food hygiene quarantine, because of highly sensitive and specific, simple, rapid and cheap. A overview of LAMP including research background, reaction principle, its application, construction method were presented in this study. The advantages and problems of LAMP were discussed, and the application was prospected.
