摘要 目的探讨EBER显色原位杂交技术(CISH)在口腔颌面部淋巴上皮癌中特有的操作与改进方法。方法应用CISH技术,荧光素标记的寡核苷酸探针EBER原位杂交检测试剂盒对40例疑为口腔颌面部淋巴上皮癌的病例进行EBER检测。结果 EBER阳性率为95%;阳性信号主要定位在肿瘤上皮细胞巢的细胞核上,呈棕黄色,周围淋巴细胞及正常组织未见明显着色;信号定位准确,染色结果清晰易辨,无背景干扰。结论简化操作EBER原位杂交检测试剂盒能够节省时间、增强信号强度和提高工作效率等。 Objective To explore the specific procedure and improvement method of EBER chromogenic in situ hybridization(CISH) technology in lymphoepithelial carcinoma of oromaxillo-facial region. Methods CISH technology and fluorescein-labeled oligonucleotide probe EBER in situ hybridization assay kit was applied,and 40 suspected patients with lymphoepithelial carcinoma of oromaxillo-facial region were given EBER testing. Results The EBER positive rate was 95%;the positive signal was mainly located at the nucleus of epithelial cell nest of carcinoma and presented as brownish yellow.Peripheral lymphocytes and normal tissues were not seen obvious coloring;the signal location was accurate,coloring results were discernible,no background interruption was detected. Conclusion A simple operation of EBER in situ hybridization assay kit(Bio Genex) is able to save time,strengthen signal intensity and enhance work efficiency.
