癫痫发病中活化小胶质细胞的参与机制 被引量:3

摘要 目的探讨分析癫痫发病中活化小胶质细胞的参与机制。方法将所选取的大鼠分为美满霉素预处理+海人酸组(MC+KA组)、单纯美满霉素组(MC组)、海人酸致痫组(KA组)和生理盐水对照组(NS组)4组。采用免疫组化法观察4个小组大鼠建模后脑内caspas-3、OX-42、c-fos免疫反应性或采用RT-PCR法检测造模后caspas-3蛋白表达量和Apaf-1m RNA含量。结果在海人酸致痫后2 h后,有明显的小胶质细胞活化、增殖出现于大脑皮质及海马部位,与此同时,c-fos蛋白的表达也明显强于对照组,上述效应在经MC干预后得到明显减弱。在建模后,caspas-3蛋白含量、Apaf-1m RNA的含量在24 h内四组之间差异无明显,而KA组在24 h、48 h时间点则高于对照组,两组差异对比有意义(P<0.05)。结论活化小胶质细胞在癫痫发病中发挥了重要作用,对进一步研究癫痫发病机制提供了全新的思路。其中美满霉素MC可能主要通过对小胶质细胞的活化的抑制,减轻神经元凋亡,提高了神经元的兴奋性,发挥抗癫痫的深井保护作用。
