丹皮酚对佐剂性关节炎大鼠IL-1β、TNF-α的影响

目的:对患有类风湿性关节炎(RA)的实验大鼠应用丹皮酚,并检测治疗后实验大鼠IL-1β、TNF-α水平,得出丹皮酚对RA的作用,探讨丹皮酚针对RA的作用机制。方法:采用弗式完全佐剂的实验方法,制备佐剂性类风湿性关节炎(AA)的大鼠模型,一周后对实验大鼠应用低剂量丹皮酚(8mg/kg)、以及高剂量丹皮酚(32mg/kg)每日1次,一共应用21d。治疗后,应用了丹皮酚的RA大鼠对足肿胀有一定的抑制作用,采用ELISA方法,得出RA实验大鼠IL-1β、TNF-α的水平。结果:和正常组比较,模型组在造模15d后立即出现了后肢的足肿胀,关节炎指数升高(P<0.05);实验显示丹皮酚很明显使实验大鼠的对侧足的肿胀时间延后,并减轻RA实验大鼠足的肿胀度(P<0.05),并且高剂量丹皮酚可以减少IL-1β、TNF-α水平(P<0.05)。结论:对患有RA的实验大鼠应用丹皮酚可以降低IL-1β和TNF-α的水平,提示丹皮酚可以抑制RA的发展。

Methods： Experimental Methods of Freund＇s complete adjuvant was usd to make the rat model of adjuvant arthritis（ AA）,A week later,rats were given on low- dose rats Paeonol（ 8mg / kg）,and high doses of paeonol（ 32 mg / kg） once daily for 21 days,For those RA rats whoe applicationed of RA Paeonol,their foot swelling were inhibited to some in rats a certain extent,by The method of ELISA was used to work out,come RA rats IL- 1β,TNF- α levels of RA ratsin. Results： Compared with the normal group,rats in model group appeareds hind paw swelling immediately duringin the modeling of the 15 days immediately hind paw swelling,and arthritis index increased（ P〈 0. 05）. Experiments showed that Paeonol obviously makes rats contralateral foot swelling time delay after RA and reduced experimental rat paw swelling（ P〈 0. 05）. and high doses of paeonol can reduce IL- 1β,TNF- α levels（ P〈 0. 05）. Conclusion： The application of Paeonol on rats with RA can reduce the level of application Paeonol IL- 1β and TNF- α,suggesting Paeonol can inhibit the development of RA.
