龙葵毛状根诱导及其抑菌活性的研究 被引量:3

摘要 [目的]探讨龙葵毛状根作为新的抑菌剂的可能性,为其开发利用提供理论依据。[方法]以龙葵叶片为试验材料,利用发根农杆菌的遗传转化技术,对龙葵叶片进行毛状根的诱导。并采用纸片法用龙葵毛状根水提物对大肠杆菌、金黄色葡萄球菌和芽孢杆菌进行抑菌活性试验。[结果]龙葵叶片经农杆菌A4侵染后7 d产生毛状根,毛状根具有多分枝、生长迅速且无向地性等特点,PCR及高压纸电泳检测结果表明Ri质粒的t DNA已整合进毛状根中。相同质量浓度下的龙葵毛状根水提物对金黄色葡萄球菌和大肠杆菌的抑菌作用较强,龙葵毛状根水提物的抑菌效果优于龙葵实生根并具有显著性差异。[结论]龙葵毛状根对金黄色葡萄球菌、大肠杆菌和芽孢杆菌均有一定程度的抑制作用,其药用价值有待进一步研究。 [Objective]The aim was to explore the possibility of using hairy roots of Solanum Nigrum L. as new antibacterial agent.[Method]Taking Solanum nigrum L. leaves as experimental material,using the transformation technology of the Agrobacterium rhizogenes,induction of hairy roots of Solanum Nigrum L. was studied. [Result]These findings suggested that hairy roots could be initiated from the cut edges of leaf explants 7 days after inoculation with the strain of A. Rhizogenes A4. The Hairy Roots had the Characters of Much branched,grow rapidly and negative geotropism and so on. The transformation of Ri t DNA was examined through PCR and high voltagepa- pereleetrophoresis. By filter paper method using hairy roots of Solanum nigrum L. and antibacterial activity was tested against Escherichia coli,Staphylococcus aureus and Bacillus subtilis,The results indicated that the inhibitory effect to E. coli and Staphylococcus aureus was better than that to Bacillus subtilis at the same mass concentration. The antibacterial effect of Solanum nigrum L. hairy root aqueous extract of Solanum nigrum L. antibacterial effect was better than the real root with the significant differences. [Conclusion] The study provided theoretical basis for the development and utilization of hairy roots of Solanum Nigrum L.
