摘要
目的:对丁香酚进行大鼠灌胃急性毒性试验和兔完整皮肤给药刺激性试验,了解其安全性。方法:丁香酚分别以5250,4200,3360,2680和2149 mg·kg-1剂量对大鼠灌胃给药,观察2周,用Bliss法测定大鼠的LD50;以1060 mg·ml^-1和265mg·ml^-1单次涂抹兔完整皮肤,观察兔皮肤的刺激性反应。结果:丁香酚大鼠灌胃的LD50为3189.9mg·kg-1,95%可信限为(2423.1~3711.8)mg·kg-1,1060mg·ml^-1对兔完整皮肤有中度的刺激性,随浓度降低刺激性减弱。结论:丁香酚的皮肤刺激性随浓度降低而减弱。
Objective:To evaluate the acute toxicity and skin irritation of Eugenol.Methods:SD rats were administered by oral gavage with eugenol at a dose of 5250,4200,3360,2680 and 2149mg·kg-1for once,respectively.After 2weeks of clinical observation,the LD50 of eugenol on rats was determined by Bliss method.Eugenol was daubed on the intact skin of rabbit at a concentration of 1060 and 265mg·ml^-1to test whether it may induce stimulus.Results:The LD50 of eugenol on rats administered by oral gavage was 3189.9mg·kg-1,and the 95% confidence intervals were(2423.1 ~ 3711.8)mg·kg-1.1060 mg·ml^-1of eugenol showed moderate irritation to rabbit skin,and this adverse effect was reduced with the decreasing concentration of the Eugenol.Conclusion:Skin irritation effects of eugenol decrease with concentration.
